AquaAmp® Fish BOL Marina PCR Kit
The AquaAmp® Fish BOL Marina PCR Kit was developed based on the patented technology of AquaGenTech Co., Ltd. (Registration No. 10-1955124, Korean Intellectual Property Office). The kit consists of all PCR reagents (i.e., primers, DNA polymerase, dNTPs, buffer, etc.) for marine fish DNA barcoding. Only the genomic DNA of the specimens needs to be prepared for the analysis using this kit.
The PCR primer set composed of the three forward and reverse primers binds to the mitochondrial cytochrome c oxidase 1 gene (CO1), the gold standard DNA barcoding region for animals. The primer set anneals the tRNA-Tyr (trnY) and tRNA-Ser (trnS) regions located upstream and downstream of CO1, respectively, and produces a single PCR band with an amplicon size of ca. 1,500 bp after conventional PCR amplification. The primers were designed to be sufficiently conserved to anneal to nearly all marine fish species, facilitating researchers to produce clear single PCR bands with a remarkably higher success rate than those designed by Ivanova et al. (2007). The kit can be easily applied to DNA barcoding of fresh fish (eggs, juveniles, and adult tissues) and processed food specimens. In addition, researchers conveniently performed DNA sequencing using specific adapters attached to the 5′ terminals of both forward and reverse primers.
AquaAmp®Fish BOL Marina PCR Kit
The performance of AquaAmp® Fish BOL Marina PCR Kit was verified using 27 marine fresh and dried fish specimens belonging to diverse taxonomic groups (i.e., Rajiformes, Acipenseriformes, Anguilliformes, Clupeiformes, Salmoniformes, Gadiformes, Lophiiformes, Gasterosteiformes, Perciformes, Pleuronectiformes, Tetraodontiformes, and Scorpaeniformes) and processed food specimens (salted and canned foods). All specimens produced clear single PCR bands with the expected amplicon sizes.
